NMN vs. NR: Which NAD+ Booster Should You Take?

The two most popular NAD+ precursors, what's the actual difference, and which is better?

NAD+ is the coenzyme that powers mitochondrial energy, DNA repair, and the sirtuins, the longevity-relevant 'genome maintenance' enzymes. Its levels drop roughly 50% between age 40 and 60, which is why NAD+ precursors are one of the hottest categories in longevity supplementation.

Two compounds dominate the market: NMN (nicotinamide mononucleotide) and NR (nicotinamide riboside). They both raise NAD+. They're not the same.

How they work

Both are precursors that get converted to NAD+ inside cells. They share most of the pathway, but:

  • NMN is one step closer to NAD+ in the synthesis pathway
  • NR has been studied more extensively in humans
  • Both reliably raise blood NAD+ at adequate doses

Evidence

NR (NIAGEN)

Over 20 published human trials, including phase 2 RCTs. Robust dose-response data, well-established safety profile. NIAGEN is the trade name and is GRAS-affirmed by the FDA.

NMN

Fewer human trials, but the ones available consistently show NAD+ elevation and modest functional benefits (walking endurance, insulin sensitivity). Most popularized by Dr. David Sinclair of Harvard. The FDA has flagged NMN's status as a dietary supplement; regulatory standing is in flux.

Which should you take?

  • Pick NR if you want the most-validated option with the strongest regulatory standing
  • Pick NMN if you want the more upstream precursor, often at lower cost per gram
  • Either way: pair with TMG to replace methyl groups consumed by NAD+ synthesis

Don't take both

Stacking NMN with NR is redundant, they end up at the same place. Choose one, dose adequately, and observe.

What about niacin or niacinamide?

They also raise NAD+, much more cheaply. The catch: niacin causes flushing at effective doses, and niacinamide may inhibit sirtuins at high doses (counterproductive for longevity goals). NMN/NR avoid both downsides.

Realistic expectations

Don't expect dramatic immediate results. Most users report subtle improvements in energy and recovery over 4-8 weeks. The deeper longevity benefits are mechanistic and probabilistic, you won't 'feel' them, but the cellular repair they support compounds over years.

Stack idea
